The multisubunit NADH:ubiquinone oxidoreductase is the first enzyme complex in the electron transport chain of mitochondria. By use of chaotropic agents, complex I can be fragmented into 3 different fractions: a flavoprotein fraction, an iron-sulfur protein (IP) fraction, and a hydrophobic protein (HP) fraction. The IP fraction contains NDUFS1, NDUFS2, NDUFS3, NDUFS4, NDUFS5, NDUFS6, and NDUFA5. Subunit: Mammalian complex I is composed of 45 different subunits. Interacts with NDUFAF3. Subcellular Location: Mitochondrion inner membrane. Similarity: Belongs to the complex I 30 kDa subunit family. Paraformaldehyde-fixed, paraffin embedded (mouse cerebellum tissue); Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Antibody incubation with (NDUFS3) Polyclonal Antibody, Unconjugated (bs-3888R) at 1:400 overnight at 4°C, followed by operating according to SP Kit(Rabbit) (sp-0023) instructionsand DAB staining.
Tissue/cell: mouse heart tissue; 4% Paraformaldehyde-fixed and paraffin-embedded;
